Sažetak
Gene trap method was used to identify genes during embryo development. The gene trap vector carrying selector (neoR) and marker (lacZ) gene without their own promoters was inserted into embryonic stem (ES) cells. From a selected ES cell clone the mouse carrying the insertion vector was made. By 5'RACE technique we found that vector was integrated into Nol1 gene. Nol1 gene encodes for proliferation associated nucleolar protein p120 which was found in malignant and rapidly dividing cells. To analyze expression pattern of Nol1 gene we used histochemical staining of mouse embryos 8.5 - 15.5-days old. In cells where Nol1 gene was active it was transcribed together with lacZ gene. The product of lacZ gene ß-galactosidase was detected as it changed its substrate X-gal into blue stain. Staining of p120 protein was found all over the embryo but not in every cell during all stages of development. Staining was high in early stages (days 8.5-12.5) of development and decreased in later stages (days 13.5-15.5). We found high subcellular staining in nucleolus and low staining in nucleus and cytoplasm. The expression pattern suggests that Nol1 gene is also involved in embryo development.
