HNE produced during the lipid peroxidation process in freshwater green alga (Chlorella kessleri FOTT et NOVAK)

Algae are primary producers in freshwater and marine ecosystems, providing basis for aquatic food chain. Unicellular green alga, Chlorella kessleri FOTT et NOVAK (strain LARG/1) was used as a model in this experiment because it is generally present in freshwater environment, easy for laboratory cultivation and has a high growth rate. Preliminary research was made to detect if the product of lipid peroxidation, 4-hydroxy-2-nonenal (HNE), a reactive aldehyde that forms protein conjugates, is present in alga C. kessleri when exposed to the hydrogen peroxide-induced oxidative stress. For dot-blot analysis, monoclonal antibodies were obtained from the culture medium of the clone derived from a fusion of Sp2-Ag8 myeloma cells with B-cells of a BALBc mouse immunized with HNE-modified keyhole limpet hemocyanine. The antibody is specific for the HNE-histidine epitope in HNE-protein (peptide) conjugates. We have detected HNE-protein adducts in cell extracts of C. kessleri but the estimation of intracellular HNE concentration was not possible. Hence, further experiments should be conducted to investigate the influence of hydrogen peroxide and exposure period on the amount of formed HNE-histidine conjugates. Algae are sensitive to wide range of pollutants and could serve as bioindicators of water pollution caused by oxidative stress-inducing compounds. Using this method, accumulation and transference of this reactive aldehyde within aquatic food webs could be monitored.
