Retinoic acid mediated neuronal differentiation of embryonic stem cells is enhanced in a culture medium with low levels of fetal calf serum

Background and purpose: Neuronal cells differentiate in vitro from embryonic stem (ES) cells after attachment of embryoid bodies formed in the presence of retinoic acid. In order to increase the proportion of neurons within embryoid body outgrowths, we decreased the amount of fetal calf serum in the culture medium used for differentiation. Materials and methods: Two levels of fetal calf serum, 10% and 1%, were applied during differentiation under graded concentrations of retinoic acid. The differentiated embryoid body outgrowths were checked for presence of neuron-like cells, neurofilament and glial fibrillary acidic protein positive cells, and Pax6 expressing cells. Results: Low levels of fetal calf serum (1%) increased the proportion of neuron-like cells and Pax6 expressing cells, but only in the presence of retinoic acid. Neurofilament and glial fibrillary acidic protein positive cells were present in the differentiated colonies. Conclusions: The addition of low levels of fetal calf serum during retinoic acid mediated differentiation of ES cells increases the proportion of cells taking neuronal differentiation pathway. This can be used to study the regulation of neurogenesis in vitro and its relation to development of the nervous system in vivo.
