Pathogen - plant interaction : Phomopsis longicolla strain pathogenicity
The aim of this research was to elaborate differences among several strains of Phomopsis longicolla isolated from soybean seed samples as first phase in breeding for resistance to this pathogen. Fungi was isolated from soybean seed samples that were collected during three years from large-scale production locations in Croatia. Two methods of artificial seed inoculation were used to test the virulence of the strains. The first method consisted in the germination of inoculated seed in wet paper. The second method consisted in the germination of inoculated seed in soil. The isolation was carried out on the infected seedlings for the purpose of determining the causative agent of wilting. Virulence of investigated strains on paper in wet chamber was determined on the basis of several factors: number of rotten seeds, number of germinated seeds, seedling lenght, seedling necrosis and number of non-germinated seeds. Virulence of strains in a pot trial was determined on basis of emerged plants 10 days after inoculation. The obtained results show a differences among investigatated P. Longicolla strains. The most virulent one will be used in further testing of a new promissing soybean lines created in the frame of soybean breeding program at the Agricultural Institute Osijek (Croatia) on resistance to this pathogen through greenhouse and field artificial infection testing.
