Interactions between Bovine Serum Albumin and Poly(Allylamine Hydrochloride) ; Potentiometric and Reflectometric Study

The association of proteins with synthetic polyelectrolytes [1] was studied on the example of the interactions between bovine serum albumin (BSA) and poly(allylamine hydrochloride) (PAH). The experiments were performed both in solution and at the interface by means of potentiometric titrations and stagnation point optical reflectometry (SPOR). Special emphasis was given on the influence of salt and ionic strength on such interactions. Potentiometric titrations of protein solutions and of protein-polyelectrolyte mixtures with aqueous solutions of different alkali metal hydroxides (LiOH, NaOH and CsOH) were performed in the presence of corresponding salts (LiCl, NaCl and CsCl). It was shown that both ionic strength and type of cation influence the titration curves, i.e. protein-polyelectrolyte interactions. The other method used for investigations of BSA-PAH interactions was stagnation point optical reflectometry. It was shown earlier that SPOR could be a useful method for studying the formation of polyelectrolyte-protein multilayers [2]. In this study, we investigated the adsorption of BSA on PAH as the outermost layer of the previously formed polyelectrolyte multilayer (PAH/PSS). It was found that the concentration of BSA significantly influences the BSA adsorption process and that above certain critical protein concentration desorption of protein occurs.
