KLJUČNE RIJEČI NA IZVORNOM JEZIKU RADA KLJUČNE RIJEČI NA ENGLESKOM JEZIKU: - 2 - PRIJEPIS SAŽETKA NA IZVORNOM JEZIKU We have assessed the Immunogenicity of Aujesky,s disease vaccine (ADV) contalning a live attenuated virus (strain Barha K/61 lacking nonessentlal glycoprotein gE) for porcine peripheral blood lymphocytes (PBL). retropharingeal (RPLNL) and bronchial lymph node lymphocytes (BLNL) using Iymphocyte stimulation test. The cells were obtained from 6 noninfected 8-week-old pigs farrowed to the sows immunized with ADV. Using flow cytometric cell sorting we hawe phenotyped and quantifled T and B cell subsets In the sampled tissues. The in vitro proliferation of RPLNL and BLNL to ADV (104 TCID50 per well) was signiflcantly gigher (P < 0.05; < 0.01) than that of PHA-or PWM-stimulated (5ľg/ml) cells, respectivety. However, the responses to BaypamunR (5ľg/ml;BPM, Leverkusen, Germany), although lower than those of RPLNL and BLNL to ADV, were much stronger when proliferation of PBL was observed. Cytolytic CD8a+ T cells occur in equal proportions (30.7% in RPLN and 29.7 in BLN) in the Iymph nodes tested. The resident CD4a+helper T cells were slightly elevated (35,7% and 42,3%, respectively) as compared to CD8a+ cells, In both lymph nodes CD21+ B cells were more frequent 35,2% in RPLM and 33.1% in BLNL) than CD1+ (20.5% and 17.8%, respectively) celis. It is likey that viral envelope glycoprotein antigens, such as gC,gB,gD, and gH, stimulated in vitro proliferation of rather reglonalLymph node lymphocytes than PBL. When BPM was appied with ADV the stimulation of RPLNL and BLNL was additionally enhanced. BPM alone seems to be a potent stimulato for porchine PBL.
