Analysis of dissolved and particulate antimicrobials in wastewater treatment using liquid chromatography/tandem mass spectrometry (LC/MS/MS)

There have been recently several reports in the literature on the removal of different classes of antimicrobials during wastewater treatment, however most of them were focused on their microbial elimination from the dissolved phase, while data on the possible role of partitioning processes are often missing. The reason for such a situation is the fact that fully validated methods for the analyses of complex sludge matrices are still relatively scarce. In order to fill this gap, a comprehensive method for the determination of both dissolved and particulate sulfonamide, fluoroquinolone and macrolide antimicrobials in wastewater and sewage sludge has been developed and validated. The dissolved antimicrobials were enriched following the most common approach, involving solid-phase extraction on hydrophilic- lipophilic balance (HLB) cartridges, while for the particulate fraction three different extraction techniques were evaluated: ultrasonication, Soxhlet and accelerated solvent extraction. After the enrichment step, the extracts of the particulate fractions were additionally purified using solid-phase extraction on HLB cartridges. The final extracts were analysed by reversed-phase liquid chromatography coupled to electrospray ionisation tandem mass spectrometry (ESI-MS/MS). The detection of all target compounds was performed in positive mode using multiple reaction monitoring (MRM). Quantification of analytes was performed using several surrogate standards, which are all added prior to the extraction step, to compensate any analyte loss during extraction and extract clean-up. The method was applied for the analysis of real municipal wastewater samples as well as for the model samples from the pilot membrane biological reactor.
