Radiation chemistry principles of radiation sterilization of drugs
Only terminal sterilization by irradiation guarantees a complete sterility assurance of drugs. Therefore radiation sterilization of drugs has evolved as a viable alternative to aseptic packaging. The research over the past 60 years has provided detailed insights into radiation action on many individual active pharmaceutical ingredients (API). However, novel concept of parentheral drug administration, enabling controlled drug delivery (CDD) and/or controlled drug release (CDR) in situ, have emphasized the need for the understanding of radiation action not only on APIs but also on their carriers and the functioning of the integral CDD/CDR systems. This work shall briefly review radiation chemistry principles encountered in radiation sterilization of APIs and CDD/CDR systems investigated over the past 15 years. Radiation chemistry data required for the assessment of radiation action on both APIs and their respective carriers, as well as on the integral CDD/CDR systems shall be discussed. Detection of irradiated drugs shall also be considered. Some emerging pharmaceutical products and processes and the opportunities for the application of radiation sterilization shall also be discussed.
