Effect of BMP-2 on leukemic NB-4 cell line
Acute promyelocytic leukemia (APL) is characterized by the clonal expansion of leukemic blasts blocked at the promyelocytic stage of myeloid differentiation. Treatment with all-trans-retinoic acid (ATRA) overcomes this maturation arrest and induces the differentiation of APL progenitors coupled with loss of their replicative potential. The treatment of NB4 cells with ATRA induced myeloid differentiation as evidenced by increased surface expression of CD11b with the major effect 72 hours after treatment. In contrast, BMP-2 did not increase the expression of CD11b when used alone. However, combined treatment with ATRA and BMP-2 significantly blocked the up-regulation of CD11b expression as the marker of myeloid differentiation that was seen in response to ATRA alone
