Evalutaion of genetic damage in workers employed in tobacco industry using the alkaline comet

Cigarette manufactores have acknowledged the medical and scientific consensus that smoking causes serious diseases such as respiratory diesease, heart disease as well as lung cancer. The aim of this study was to find the possible genotoxic effects of occuaptional exposure to tobacco dust and additionally exposure to a source of strontium-90.All togethor 60 subjects (20 exposed and 40 controls) participated in the study.The assesssment of primary DNA damage in peripheral blood leukocytes was performed using the alkaline comet assay. Tail lenght was measured as main comet parameter. The results showed that the comet tail lenght measured in exposed workers were in the range of mean values 12.52+0.12um to 15.85+0.20um. In control group tail lenght mean values ranged 10.37+0.19um to 15.08+0.15um.Within the exposed population, there were no significant inter-individual differences in DNA damage found. Cigarette smoking as well as age and sex was not a potent confounding factor in control and exposed subjects. The observed non-significant differences in length mean of both groups suggest that tobacco dust and strontium-90 does not significantly contribute to DNA damage. As a very sensitive and rapid method for assessment of genome damage, the alkaline comet assay pointed to the importance of biological indicators in providing information on the actual risk of tobacco dust and strontium-90 exposed individuals.
