ADME data for polyphenols characterized by reversed-phase thin-layer chromatography
The chromatographic behavior of polyphenols (flavonoids and phenolic acids) have been investigated by reversed-phase thin-layer chromatography (RP-TLC), in order to establish relationship between chromatographic indices and selected ADME (Absorption, Distribution, Metabolism and Elimination) parameters. Good correlation between chromatographic index RM (a measure of the solute`s interaction with the lipophilic, stationary phase on the one hand and the hydrophilic, mobile phase on the other hand) and concentration of buffer in the mobile phase (r ≥ 0.994) might be taken as a measure for the suitability of examined system for polyphenols’ lipophilicity estimation. Experimentally obtained chromatographic indices (RMw - a measure of the solute`s interaction with the lipophilic, stationary phase and pure water, as the mobile phase and φ 0 - the volume fraction of the organic modifier/buffer in the mobile phase for which retention is zero) were compared to ADME parameters calculated by software packages based on different theoretical approaches. According to our results, we could establish the correlation between chromatographic behavior and lipophilicity, solubility, plasma-protein binding and oral absorption of investigated polyphenols.
