BMP-7 is expressed in colon tissue during experimental inflammatory bowel disease

Bone morphogenetic protein-7 (BMP-7) is member of the transforming growth factor-beta superfamily, and plays an important role in the development and healing of numerous tissues, especially bone. The findings of presence BMP expression in developing intestine and mutations of BMP Ia receptor as well as Smad 4 in colon disease suggest on important role for BMPs in the intestine inflammatory and cancer diseases. Using RT-PCR, we have previously shown the expression of BMP-7 in colon tissue in both, acute and chronic phases of experimental inflammatory bowel disease. In addition, BMP-7 treatment decreased the expression of BMP-7 in the intestine. Here, we report the immunohistichemical expresson of BMP-7 in colon to show precise localization of BMP-7 binding in diseased and BMP-7 treated animals. We investigated the expression of BMP-7 in rat colon during different phases of inflammatory bowel disease by assessing tissue samples of 2, 5, 10, 14, and 30 days of colitis with immunohistochemistry. We also explored the effect of therapeutical and prophylactical BMP-7 treatment on BMP-7 expression in colon samples. We found strong staining with BMP-7 antibody in rat colon samples in acute as well as chronic phases of disease. The BMP-7 expression was predominately located in mucosa, in crypt colonocytes and in lamina propria. In acute phase of colitis (2, 5, and 10 days after induction colitis) the highest BMP-7 expression was found in region of ulcus while in chronic phase of colitis (14 and 30 days after induction colitis) the expression was mostly placed in the colonocytes along the crypts. BMP-7 treatment and pretreatment significantly reduced its expression in colon tissue and was found in fewer crypt colonocytes. These immunohistochemical dates confirmed our previous RT-PCR data and suggest that BMP-7 acts as an antiinflammatory agent involving and affected predominately cells of colonic mucosa.
