In vitro susceptibility testing of Malassezia pachydermatis to miconazole and clotrimazole

Malassezia pachydermatis is an opportunistic pathogen of the external ear canal of dogs. Regarding special clinical conditions, mainly in cases of therapeutical failure related to external otitis, additional efforts have been made to perform susceptibility testing. Most comercial products for topical treatment of otitis externa in dogs contain clotrimazole or miconazole as antifungal agents. 20 strains of M. pachydermatis isolated from dogs with symptoms of otitis externa were investigated for in vitro susceptibility to various concentrations (0.03-64 μ g/ml) of two drugs, using broth microdilution method. Sabouraud dextrose broth supplemented with 1% Tween 80 was used for the culture and for ten-fold dilutions for the inocula. The ranges of minimal inhibitory concentrations, determined after 48 hours, for the 20 isolates were: a) clotrimazole, from 0, 5 μ g/ml to 64 μ g/ml (MIC50 8 and MIC90 64 ) and b) miconazole, from 0, 25 μ g/ml to 64 μ g/ml (MIC50 2 and MIC90 64). The isolates of M. pachydermatis showed great variations in MICs to these agents. That could be due to broad use of these antifungals or selection of strains with higher MICs related to prolonged antifungal treatment of dogs from whom more resistant strains were isolated. The use of this method allows the assessment of the MICs of large number of isolates to antifungals. Procedure is easy to perform and inexpensive compared to other tests which makes this a method of choice for laboratory use.
