Development of Practical and Accurate HPLC Methods for Enantioselective Analysis of Fluoxetine and Citalopram
Simple and robust HPLC methods for the direct separation of enantiomers of the popular antidepressive medicines fluoxetine and citalopram were developed. The enantioseparation was tested on numerous commercial chiral HPLC columns. The mobile phase compositions were systematically studied to obtain the optimal chromatographic methods. For fluoxetine the most convenient separation was determined on amylose Chiralpak AD column, and for citalopram on cellulose Chiralcel OD-H column. Validation of methods in selected conditions has shown that the chosen methods are selective and precise with linear response of detector for the both pairs of enantiomers.
