Growth promoting effect of human plasma ultrafiltrate bioactive fraction on human non-functioning pituitary adenoma cells in vitro

There is increasing evidence for the role of certain neuroendocrine and/or systemic humoral factors in pituitary differentiated function and tumorigenesis. Therefore the effect of the human plasma ultrafiltrate bioactive fraction (tentatively termed as TBP) on growth of 8 non-functioning adenoma cell cultures was investigated. TBP was prepared by anion-exchange chromatography from the pooled EDTA-plasma of 49 healthy, male blood donors. Samples of adenoma tissue, obtained at transphenoidal microsurgery, did not contain pituitary gland tissue in addition to adenomatous tissue (as revealed by histology). Sliced adenoma tissue fragments were dispersed mechanically. Cells were cultured in 96-well culture plates (2 x 100 000 cells in 1 ml of culture medium) containing DMEM supplemented with 10 percent fetal calf serum, penicilline and streptomycin at 37C and 5 percent CO2 for 4 days. The culture medium of the cell cultures treated with TBP contained the sample of bioactive plasma ultrafiltrate fraction at 100g/l plasma equivalent. Cell growth was determined by 3H-thimidine assay, indicating DNA synthesis, for triplicates of cultures. In the presence of TBP DNA synthesis was doubled (on the average 197.8 percent of respective control +-51.5) (p less than 0.05). While such promoting effect was found in 5 out of 8 adenoma cell cultures, none of the remaining adenoma cultures were inhibited. From these results, it appears that TBP could be one of the humoral factors involved in the regulation of proliferation of pituitary adenoma tissue.
