Influence of organic selenium (Sel-Plex®) supplementation on age-related changes in antioxidant system of tight muscle of broiler chickens

The influence of organic selenium food supplements on the activity levels of antioxidant enzymes, reduced glutathione (GSH) concentrations and lipid peroxidation in tight muscle (M. biceps femoralis) of Ross heavy line chickens were investigated. At the age of 2, 4 and 6 weeks, 10 chickens fed a standard diet (control containing 0.15 ppm selenium (Se) as sodium selenite) and 10 fed the same diet to which 0.3 ppm organic Se as Sel-Plex® ; ; ; (Alltech Inc.)were slaughtered. Thigh muscle homogenates were analysed for GSH-Px activity, copper/zinc superoxide dismutase (CuZnSOD), manganese superoxide dismutase (MnSOD) activities and glutathione (GSH) and lipid peroxide (TBARS) concentrations.A clear trend toward increasing thigh muscle GSH-Px activities in experimental chickens from 2 to 6 wks of age was measured (P<0.01), whereas in 6 wk old control chickens increased activities compared with 2- (P<0.05) and 4-wk-old (P<0.001) chickens was observed. Sel-Plex® ; ; supplementation resulted in significantly higher GSH-Px activity in thigh muscle of broilers at 4 and 6 wks of age in comparison to the control group (P<0.01). The CuZnSOD activity decreased in 6-wk-old control chickens compared with 2- (P<0.01) and 4-wk-old (P<0.05) chickens. Tigh muscle from chickens 4 wks af age given Se showed lower CuZnSOD activity than 2- and 6-wk-old chickens (P<0.05). The CuZnSOD thigh muscle activity was significantly lower in 4-wk-old experimental chickens than in controls (P<0.001) due to intensive metabolism and higher body weight gain in experimental chickens in that period. The MnSOD activity in both control and experimental chickens reached its lowest (P<0.05) level at 6 wks of age. Thigh muscle from chickens fed Sel-Plex® ; ; showed higher MnSOD activity (P<0.01) at 2 wks of age. The GSH concentration decreased (P<0.01)in thigh muscle of Sel-Plex® ; ; chickens as the chickens grew older. In 6-wk-old chickens GSH concentration showed a significant decline compared with 2- and 4-wk-old chickens (P<0.001). Thigh muscle from 4-wk-old chickens given Sel-Plex® ; ; showed lower GSH concentrations (P<0.01) compared to those fed the standard diet. The results of this study demonstrate that the organic selenium food supplement had a positive effect on broiler chicken GSH-Px activity in thigh muscle.
