Quantitative in vitro Study of Semaphorin5A Functional Role in Neocortical Axons Growth in Mouse
Slice culture methods and functional assay were used to study possible influence of signal glycoprotein semaphorin5A on the development of cortical efferents in mouse during late embryonic period (E15-E18). Semaphorin5A is mainly expressed in central nervous system. Most members of semaphorins family show influence on axonal outgrowth, elongation, and collateralization or steer them in the developing periphery and central nervous system. This study evaluates influence of semaphorin5A on growing neocortical axons. Sema5A promotes their outgrowth and elongation when is expressed uniformly on growing carpet in vitro. Under those conditions, during 72 hour’ s examination period, semaphorin5A do not steer axons or modulate collateralization. We conclude that semaphorin5A can join “ wiring molecules” which serve as signals for assembling the intricate network of cortical connections.
