Arteriolar responses to dilator agents and elevated PO2 in consomic SS.BN13 rats
Resistance vessels of hypertensive Dahl salt sensitive (Dahl S) rats on high salt (HS) diet exhibit an impaired relaxation to hypoxia (Microcirculation 8:115-27, 2001), acetylcholine, and iloprost ; and an enhanced sensitivity to elevated PO2 (Int. J. Microcirc.: Clin & Exp. 13:83-89, 1993). Consomic SS.BN13 rats are an inbred strain with chromosome 13 from Brown-Norway rat introgressed into the Dahl S genetic background. The aim of this study was to define the responses of arterioles to vasodilator stimuli and elevated PO2 in the cremaster muscle of consomic SS.BN13 rats on short term low salt (LS) (0.4% NaCl) and HS (4% NaCl) diet. Third order cremasteric arterioles of SS.BN13 rats exhibited a significant dilation in response to acetylcholine and sodium nitroprusside, and a significant constriction in response to superfusion with both 5% and 21% O2 solution. Dilator responses of arterioles of SS.BN13 rats tended to be larger than those of Dahl S rats, consistent with the hypothesis that substitution of chromosome 13 restores vascular relaxation mechanisms that are impaired in Dahl S rats.
