Phospholipids of the Mitochondria of Recycled Brewer's Yeast
Bottom fermenting brewer's yeast is used traditionally for the production of lager beer. Aerobically grown pure culture of the brewer's yeast undergoes changes of its physiological properties during the fermentative cycle in anaerobic process of alcoholic fermentation. It is the main reason why in industrial beer production up to the fifth generation of yeast cells are usually used. Mitochondria are the central metabolic organelles and their functional state plays a critical role in brewing yeast physiology and fermentation performance. It is well established that ethanol tolerance of brewer’ s yeast is closely tied to its lipid composition. In this work phospholipid composition of the mitochondria of recycled brewer's yeast (the second generation) from Buzet brewery "BUP" has been analyzed. Mitochondria were isolated from the yeast cells after enzymatic disruption of the cell wall. Individual phospholipid classes were separated from total lipids by two-dimensional thin-layer chromatography and quantified spectrophotometrically as inorganic phosphorus. The content of total phospholipids in the mitochondria expressed vs. proteins accounted for 0, 15 mg/mg. Major phospholipid classes were phosphatidylcholine (29, 0% of total phospholipids), phosphatidylinositol (26, 5%) and phosphatidylethanolamine (19, 1%). Cardiolipin, typical mitochondrial phospholipid, made up 6, 0%. Data obtained indicated that ethanol, which accumulated during fermentation, and anaerobic conditions both affected mitochondrial phospholipid composition.
