Characterization of the proteolytic enzyme - keratinase
Keratinases are proteolytic enzymes that catalyze degradation of keratin. Preparations of the keratinase have different characteristics, because they could be produced by the different sources. They have molecular weights in the range of 16 - 440 kDa. Most of the keratinases have temperature optimum in the range of 25-40 C. The keratinase are stable at temperature under - 20C. They have the optimal activity in the range of pH 7-9, except the acid keratinases which have optimal pH between 2.5-4.5. Most of the purified keratinases hydrolyze a wide range of proteins, such a bovine serum albumin, casein, hemoglobin, etc. After the fermentation of the selected fungi the crude keratinase was prepared by ultrafiltration and lyophilization. It was purified by hydrophobic interaction chromatography followed by ion-exchange chromatography. The purity and the molecular weight of the enzyme were detected by SDS-PAGE electrophoresis. The keratinase was partially characterized and compared with proteinase K and thermolysin. The pH and temperature optimum were determined by various test activities. The effects of the temperature and the concentration of the enzyme on the volume activity of keratinase was studied.
