Identification of T lymphocite subpopulations during allograft rejection
The mononuclear cell infiltrate s in human renal and rat skin allografts were examined by monoclonal antibodies using an avidin-biotin-peroxidase technique. Human renal biopsies were obtasined from patients during rejection chrisis, while rat skin allografts were taken and analysed aproximately at the time when skin grafts were undergoing rejection. A great proportion of infiltrating cells in renal and skin allografts was stained with monoclonal antibodies that identify T lymphocites. Using monoclonal antibodies specific to CD4 (T helper/inducer)and CD8 (cytotoxic) surface markers, we were able to estimate the relative proportion of these cells infiltrating the skin allografts in rats as well ashuman renal allografts. By using this method it is possible to estimate the allograft rejection before the clinical signs of rejection and the occurence of deteritoriation of graft functions.
