Technical parameters and normal values of the rosette assay with lymphocytes from human peripheral blood
A method for detecting rosette forming cells in human peripheral blood is described. Two types of rosette forming cells were determined: spontaneous rosettes (sRFC), detected when lymphocytes attach sheep red blood cells (SRBC), and induced rosettes (iRFC), detected when lymphocytes attach SRBC pretreated either with anti-SRBC antibodies (EA-RFC) or with anti-SRBC antibodies and complement (EAC-RFC). Technical parameters of the test were defined, such as procurement of the cells for testing, and temperature and duration of incubation. The values of sRFC and iRFC in the peripheral blood of twenty-nine healthy persons were determined. The values obtained for sRFC ranged from 17.9 % to 50.1 % and the values for EA and EAC-RFC from 6.4 % up to 19.9 % or from 6.0 % up to 20.5 %, respectively.
