Alkaline phosphatase and 5-nucleotidase as indicators of zinc status in swine
The aim of the study was indirect detection of zinc deficiency in swine by determination of catalytic activites of zinc-dependent enzymes. The study included 42 German Landrace x Pietren x Large White x Swedish Landrace weaned crossbreds divided into one control group and two experimental groups (T1, T2) of 14 animals each. Control group animals (C) received insufficient dietary zinc, experimental group T1 received zinc supplementation in the form of ZnSO4 (80 mg/kg feed) and experimental group T2 received zinc supplementation in the form of zinc methionate (40 mg/kg feed). Blood samples were collected on days 1, 28, 56 and 105. Zinc concentration, total alkaline phosphatase catalytic activity and alkaline phosphatase isoenzymes were determined in blood serum, and 5-nucleotidase activity was determined in isolated lymphocytes. A decreased zinc concentration in animal serum correlated well with the decreased total alkaline phosphatase activity in serum (r=0.94) and the decreased 5- nucleotidase activity in isolated lymphocytes (r=0.67). Decreased zinc concentrations most probably reflect changes in the relative proportion of alkaline phosphatase isoenzymes. Based on the good correlation between the changes of zinc concentration in serum and activities of zinc-dependent enzymes tested, we propose the use of reliable, rapid and simple analytical procedures for determination of alkaline phosphatase activity in serum or of 5-nucleotidase activity in animal lymphocytes for the assessment of zinc deficiency instead of expensive and complex atomic absorption spectrophotometry.
