Sažetak
The urokinase-type plasminogen activator (uPA) and plasminogen activator inhibitor-1 (PAI-1) have a major function in tumor invasion and metastasis. Higher levels of these proteases are linked to poor prognosis in variety of malignant tumors. uPA and PAI-1 were expressed in most thyroid carcinomas, however no relationship between their expression and clinicopathological parameters could be determined. Objective: Determination of the uPA and PAI-1 cytosol concentrations in thyroid tumors and correlation to known prognostic factors. Design: uPA and PAI-1 concentrations were measured using enzyme-linked immunosorbent assay in cytosol samples of thyroid tumor and normal tissue of 23 patients. Results: Both uPA and PAI-1 concentrations were significantly higher in thyroid cancer than in normal tissues. There was a positive correlation between uPA and PAI-1 concentrations in malignant tumors. Both uPA and PAI-1 levels were associated with tumor differentiation, size, multicentricity, extrathyroidal invasion, and presence of distant metastases. The uPA and PAI-1 levels showed significant difference among histological types of thyroid cancers. Values of PAI-1, but not uPA, were significantly higher if lymph node metastasis were present. Conclusion: These data suggest that uPA and PAI-1 are functionally active in thyroid cancer and are associated with certain prognostic parameters.
