Calmodulin Kinase II Constitutively Binds, Phosphorylates, and Inhibits Brush Border Na+/H+ Exchanger 3 (NHE3) by a NHERF2 Protein-dependent Process
1) NHE3 is a CaM kinase II and Ca2+/CaM binding protein. 2) CaM kinase II binds to NHE3 under basal conditions, at aa 585-605. 3) CaM kinase II dependent phosphorylation of NHE3 inhibits NHE3 under basal conditions and this inhibition is mediated by NHERF2. 4) Ca2+/CaM also binds NHE3 under basal conditions at aa 585-605. CaM kinase II and Ca2+/CaM are part of the physiologic NHE3 regulation that occurs in fibroblasts as well as in BB of ileal Na absorptive cells.
