Different expression of integrins and ICAMs in peripheral blood, lymph nodes and bone marrow in B-CLL
B-CLL patients have variable tumor mass distribution within major cell compartments, and as a consequence different clinical presentation. To evaluate role of integrins and their ligands ICAMs in homing of B-CLL lymphocytes to various lymphoid compartments we have evaluated the expression of integrins (CD11a, CD11b, CD11c, CD18, CD29, CD49c, CD49d and CD61) and ICAMs (CD54, CD102 and CD50) in 33 B-CLL patients in samples taken from peripheral blood (PB), bone marrow (BM) and lymph nodes (LN) by two/three color flow cytometry (on CD19 positive cells). We found stronger expression in PB than in BM for CD11a and CD102, while CD54 was stronger in BM than in PB (p<0, 05). CD11b and CD102 were stronger in PB than in LN, contrary to CD54 (p<0, 05). CD102 was stronger in BM than in LN, contrary to CD18, CD11a and CD11c (p<0, 05). By calculating logarithmic values of MFI ratios between two compartments for respective integrin or ICAM, differences were quantified and their relationships with clinical and laboratory parameters were evaluated. We found strong positive correlations of LN to PB ratio for CD11a, CD11b and CD18 with peripheral blood lymphocytosis (p<0, 05). Lymph node size was negatively correlated with LN to PB ratio for CD11b and with BM to PB ratio for CD54 and CD61 (p<0, 05). Spleen size was negatively correlated with BM to PB ratio for CD11c and CD102 (p<0, 05). BM insufficiency was associated with significantly higher BM to PB ratio for CD18, CD11a, CD11c, CD49d, CD54 and CD102, and with lower LN to PB ratio for CD102 (p<0, 05). This study shows that expression of selected integrins and ICAMs on B-CLL cells is significantly different among lymphoid compartments. Multiple associations with clinical and/or laboratory parameters with quantified differences in integrin and ICAM expression between compartments indicate their possible role in resulting clinical presentation. Taking together, adopted investigation approach proved feasible, disclosed yet unexplained interesting interactions and warrants further studies.
