mutT - Independent GASP Phenomenon of Escherichia coli

Famine and feast are the most common lifestyle of the microbial cells. In their natural environment bacteria spend the majority of their time in the stressful conditions where nutrients are scarce. Highly dynamic starved cultures of Escherichia coli have evolved through mutations and evolutionary selection mechanisms that enable them to survive. The aim of this study was to evaluate the interaction between two species of Enterobacteriaceae: E. coli and Salmonella enterica serovar Typhimurium. Growth advantage in stationary phase (GASP) is a mechanism which allows bacteria to survive during prolonged carbon starvation stress, and through strong GASP phenotype an old minority bacterial population grows and invades a young majority population under this stressful condition. We observed the dynamics of growth of mutant E. coli mutT, in separate cultures as well as in the mixed ones. Mixed cultures consisted of 10-day-old culture of E. coli mutT TetR, and young, 1-day-old, wild type bacteria S. enterica StrR or E. coli StrR, in different ratio of 1:10 and 1:100, respectively. Results indicate that bacteria E. coli mutT exhibits weak GASP phenotype in the rich medium (LB), contrary to the minimal medium (M9), the same mutant shows strong GASP phenotype in the mixed cultures with wild type bacteria S. enterica or E. coli.
