Regio and chemoselective transformations of saccharides catalyzed bz esterases isolated from mammalian sera
Mammalian sera are rich sources of esterase activities with specificity for carbohydrate esters as will be demonstrated for enzymes isolated from rabbit and guinea-pig sera. Individual enzyme fractions were isolated and separated from sera by column chromatography. They were characterized by using series of substrates and inhibitors specific for various possible types of esterases usually present in mammalian sera. Kinetic studies were also undertaken with radiolabelled monosaccharides as substrates. Separated enzyme fractions were then applied in regio and chemoselective deacylations of acylated monosaccharides. Acyls used were acetyls, pivaloyls and N-acylaminoacyls. The degree of regioselectivity and the direction of hydrolysis depend on the sugar structure, enzyme source, and the particular characteristics of each isolated fraction. Furthermore, high degree of chemoselectivity was observed in the case of compounds bearing structurally different acyl groups.
