Summary of annual progress report of Partner 3 in WP3.B Availability and impact of pollutants on biota Partner 3 Rudjer Boskovic Institute Division for Marine and Environmental Research Bijenicka c. 54, HR-10000 Zagreb, Croatia 1) PLANNED ACTIVITIES WITHIN THE SARIB PROJECT FOR THE FIRST 12 MONTHS a) To implement multidisciplinary study for correlating subtoxic exposure and/or effects of selected priority pollutants to health status of feral fish (European chub) b) To combine chemical, biological, veterinary, biochemical and ecotoxicological aspects in assessing the impact of metals and specific organic pollutants on the health of indigenous fish species from Sava River. c) To define health status of feral fish (European chub) by its examination on parasites, viruses, specific bacteria, accompanied with histological analysis of fish tissue. d) To determine biomarkers and effect endpoints (metallothioneins, EROD activity, AMES test) in target detoxification organ such as liver to assess potential effects of pollutants on feral fish (European chub) health. e) To determine metals (Zn, Cu, Cd) known as inducers of metallothioneins in fish liver cytosol. f) To determine PAHs and PCBs in liver tissue of European chub from Sava River. g) To accomplish the deployment of “ passive samplers” as time-integrated metal concentrations by means of Diffusive Gradients in Thin films (DGT) and respectively selected organic pollutant (PAHs, PCBs) concentrations by means of Semi-Permeable Membrane Devices, SPMDs at selected locations in the Sava River water column. 2) WORK DONE IN THE FIRST 12 MONTHS Planned activities were conducted as follows: a) Acceptable sampling technique according to the standard HRN EN 14011:2005, Water quality-Sampling of fish with electricity for feral fish (European chub) adopted. b) Suitable fish sampling sites and passive sampler deployment sites on river Sava selected from Slovenian-Croatian border downstream to Jasenovac. c) Analysis of chub meat on moisture, ash, protein and lipid content performed. d) Initial database on the incidence of diseased fish in the Sava River created. It includes: -virusological examination ; -bacteriological examination from gills, liver, spleen, kidney ; -ectoparasites from gills, fins and skin ; -endoparasites in internal fish organs, especially intestine ; -histological analysis of liver, kidney, spleen and intestine of European chub caught at 4 selected locations on Sava River. Bacteriological analysis of water at the site of fish sampling was accomplished, too. e) Biomarkers and effect endpoints (metallothioneins, EROD activity, AMES test) in fish liver as detoxification organ determined. Metallothioneins determined in gill cytosol, too. f) Cytosolic proteins analysed in S50 fraction of liver and gills. Metals (Zn, Cu, Cd), inducers of metallothioneins, determined in the same heat-treated cytosolic fraction of liver and gills in which metallothioneins were analysed, too. g) Fish plasma samples shipped to Partner 10 with the purpose to develop an ELISA method for vitellogenin analysis in European chub from Sava River. h) Total PCBs, sum of 7 PCBs and total DDTs determined in liver and muscle of Sava River European chub. Mass partition of organic pollutants expressed on wet tissue mass and respectively on lipid mass. i) Pilot deployment of “ passive samplers” for metals (DGT) and organic pollutants (SPMDs) performed at one Sava River location. j) Method of metal extraction from passive samplers (DGT) tested in the laboratory and adopted for field work. k) Logistic of extraction of passive samplers for organic pollutants under development. 3) DEVIATION FROM THE PLANNED ACTIVITIES During the first year no significant deviation from the planned activities has occurred. Due to the complexity of approach and problems that are related to the purchase and delivery of the necessary equipment, acquiring the implementation of new methodology and new technical performances, the following activities occurred slower than planned: -deployment of “ passive samplers” for metals (DGT) and organic pollutants (SPMDs). The duration of deployment needs further site-specific evaluation. In the follow-up phase deployment will occur at all selected sites. -extraction of metals and respectively organic pollutants from passive samplers needs further validation and improvement. -determination of PAHs in fish bile in follow-up period -determination of PAHs in fish liver in follow-up period.
