In search for genotoxicity: fish
Genotoxicity monitoring of marine and freshwater environment was conducted as a part of the three-year CROWAT project. DNA damage was evaluated in erythrocytes of fish collected from their natural habitat (Mullus barbatus) as well as from caged fish (Dicentrarchus labrax and Cyprinus carpio). DNA integrity of fish blood cells was analysed by comet and micronuclei (MN) assays. While the alkaline comet assay detects primary repairable, therefore reversible DNA lesions (alkali labile sites, single strand DNA breakages), the MN assay detects more persistent DNA lesions or aneugenic effects that cannot be repaired. Persistence of MN in the cell is therefore longer than single strand DNA breakages and alkali labile sites that are being, more or less constantly repaired. The benefit of the combined use of these two assays in genotoxicity monitoring of Croatian marine and freshwater environment will be discussed.
