Granulysin expression in peripheral blood and decidua of the first trimester pregnancy

PROBLEM: Granulysin (G) is a novel cytolytic molecule expressed in natural killer (NK) and activated cytotoxic T cells. It is capable to lyse a variety of tumour cells and microbes with the ability of direct killing of intracellular pathogens after the entrance in the target cells through perforin pore. Since little is known about the G expression at the materno-fetal interface, we analysed the presence and distribution of this molecule in the first trimester pregnancy decidua and peripheral blood (PB) of pregnant women. MATERIAL AND METHODS: Phenotype of granulysin in PB of pregnant women and freshly isolated decidual mononuclear cells was analysed with triple labelling of surface (CD3 and CD56) and intracellular (granulysin) antigens using cell permeabilisation method and flow cytometry. Intracellular localisation of the granulysin in comparison with perforin was analysed with confocal microscopy. The antibodies used were: biotinilated mouse IgG1 anti- granulysin monoclonal antibody (mAb), produced by Prof. S. Saito, Toyama, Japan), mouse IgG2b anti-perforin mAb, IgG1, anti-CD3-FITC, IgG1 anti-CD56-cy or isotype matched control (all from Becton-Dickinson, Erembodegen, Belgium) RESULTS: The percentage of granulysin positive (G+) cells in PB of pregnant women is about 10% and it is distributed 70% in T cells and 30% in NK cells. Contrary to PB, decidua contains 20% of G+ cells and the majority of G is expressed in NK cells (70%). Only about 10% of T cells are G+. The frequency of G within decidual NK cells is 70%, while 10% of PB NK cells are G+. The most of G is stored in cytoplasmic granules near the cell membrane. Granulysin rarely colocalizes with perforin which is mostly in centrally positioned granules. The most of the decidual lymphocytes are both G+ and perforin+. CONCLUSION: Beside perforin, granulysin seems to be the second abundant cytolytic molecule expressed at materno-fetal interface.
