Immunological role of Tumor Associated Glycoprotein-72
Problem: The distribution of the mannose receptor (MR) in the first trimester pregnancy decidua and term placenta, possible binding properties and the effect of Tumor Associated Glycoprotein-72 (TAG-72) to the MR and tolerogenic program in early decidual MR positive cells were analysed. Methods of study: Distribution of MR was analysed by immunocyto/histology and immunofluorescency. Endocytic activity was measured by FITC-Dextran uptake in untreated macrophages or macrophages treated with mannan or TAG-72. The interleukin-4 protein expression was analyzed by immunofluorescency in permeabilized cord blood T cells after 3 days-culture with magnetically separated decidual CD14 positive cells untreated or pretreated with TAG-72, LPS or TAG-72 and LPS. Results: MR- positive cells were disseminated in term placenta, mostly concentrated around the early decidual glands. Aproximatelly half of CD14-positive cells were MR-positive, which if freshly isolated internalized FITC-Dextran more efficiently than 18-hour cultured and mannan or TAG-72 pretreated freshly isolated cells. Decidual CD14-positive cells pretreated with TAG-72 or TAG-72 and LPS enhanced interleukin-4 synthesis in cord blood T cells. Conclusions: The mannose receptor positive macrophages surrounding early decidual glands are capable to bind and internalize decidual secretory phase mucin TAG-72 that enables them to induce interleukin-4 production in naive T cells.
