Ig heavy gene rearrangement in lymphoid neoplasia - a molecular biology approach
We selected the appropriate and reliable molecular biology method to provide a unique marker for detecting a monoclonal B lymphocyte population, which allows us to distinguish a monoclonal versus a polyclonal rearrangement of the IgH gene in B cells. We applied a semi-nested PCR based on: - germline state (in all cells except B lineage lymphocytes) V and J segments of IgH gene are several thousands bases apart and therefore cannot be amplified by the selected primers - during B lymphocyte differentiation V and J segment are brought close together and can be amplified by PCR using specific primers The method is especially useful in cases: - when conventional methods do not give a conclusive diagnosis - when there is limited supply of tissue (MALT)
