It is known that the plant extract of Amorphophallus konjac K. Koch has the hypoglycemic effect because it could delay the glucose absorption from the intestine. Regenerative and other metabolic effects of B-complex vitamin from yeast are known as well. We supplemented the yeast (3 g/100g food), or yeast (3 g/100g food) and Konjac (600 mg/100 g food) in combination, to the food of alloxan diabetic rats. The effect of such feeding on the kidney damage, which is well known complication of diabetes, was examined after 36 days of treatment. We measured the catalytic concentration of N-acetyl-b-D-glucosaminidase (NAGA) in kidney and urine. It is an early biochemical indicator of morphological changes in diabetic kidney. The activity of alkaline phosphatase (ALP) in kidney was measured for the examination of membrane stability. The ALP activity was measured spectrophtometrically and NAGA activity spectrofluorometrically using 4-methylumbelliferyl substrates. The results show that NAGA activity in diabetic kidney increased to 190 % and in urine to 270 % as compared to healthy animals (100 %). After yeast treatment the activity partially decreased for about 50 % in comparison to diabetics fed with regular food. The addition of yeast and Konjac combination to diabetic rat food decreased NAGA activity nearly to the values of healthy rats. However, 30% decrease of soluble ALP in kidney was observed in diabetes in relation healthy rats. The yeast treatment partially increased ALP activity, while the yeast and Konjac combination completely restitute the ALP values to healthy ones. Our results show that addition of yeast and Konjac combination to diabetic rat˘s food might retard the kidney damage in our experimental conditions.
