Genetic diversity evaluation of OS-soybean germplasm using microsatellite markers

In soybean (Glycine max (L.) Merr.), genetic diversity evaluation are based on pedigree lineages, phenotypic (morphological and biochemical traits) and DNA markers.These measures together provide more reliable genetic diversity evaluation. Diversity information is of paramount importance in parental selection, crucial aspect of soybean breeding that maximizes genetic improvement. Regarding, more distantly related parents might be expected to contrast for more alleles in target traits and thus increase the probability that a breeder will select a superior recombinant among progeny which exhibits target trait. Recent years, in the frame of the soybean breeding program at the Agricultural Institute Osijek (Croatia) has initiated with application of molecular marker technique as new genetic diversity measure. The initial fingerprinting of several OS soybean genotypes has performed in collaboration with the University of Guelph (Canada) in their biomolecular laboratory using simple sequence repeats (SSR) or microsatellite markers. Selected OS genotypes were differed in wide range values of morphological and genealogical traits. Fourteen pairs of soybean primers flanking the micorsatellite regions were selected. PCR amplification was performed on each OS-soybean genotypes, using primers for each SSR locus. In this paper will be presented the obtained results of this study as well as morphological data of tested materials. Thus, combining molecular marker technique with pedigree information, phenotypic markers and statistical procedures has provided a useful tool for more accurate and complete evaluation of genetic diversity and throughout, its more effective utilisation into current soybean breeding program.
