Organic - Inorganic Nanocomposite Coatings for Biomedical Applications II

Multilayer (ML) films formed by alternated adsorption of anionic and cationic polyelectrolytes (PE) have proven to be an efficient way to prepare functionalized surfaces (Decher, Science, 277 (1997) 1232). Recently, it has been indicated that PE ML could induce calcium phosphate nucleation (Ngankam et al, J. Am. Chem. Soc., 122 (2000) 8998) and that crystalline calcium phosphate could grow on them (Gergely et al, Key Engineering Materials, 240-242 (2003) 287) . These findings prompted us to attempt to develop novel organic-inorganic coatings for artificial bone implants consisting of PE multilayers and “ in-situ” grown calcium phosphate crystals. In this work we report on preparation and characterization of novel organic-inorganic coatings for biomedical implants. PE multilayers, (PLL/PGA)nPLL, (where PLL is poly-l-lysine, PGA is poly-l-glutamic acid and n is the number of layers) were deposited on titanium plates and on top of them amorphous calcium phosphate (ACP) particles were adsorbed Using this procedure, complex structures, consisting of i PE multilayers, alternating with j layers of ACP particles (i = 1-6, j = 1-5) were prepared. After immersing such structures into dilute calcifying solutions, OCP crystals grew throughout the organic matrix, yielding organic-inorganic composite systems. It was shown that:  Surface coverage of substrate improves with increasing i and j.  Adhesion to the substrate: Coatings B > coatings A.  Proliferation of human osteoblast cells on coatings B is enhanced only when OCP crystals are covered with a PE multilayer. Advantages: the PE multilayers provide an organic matrix, into which biologically important macromolecules, such as enzymes, growth hormones and/or drugs may be incorporated.
