Comparison of 864 and 935 MHz microwave radiation effect on cell culture

The aim of our study was to evaluate and compare effect of 864 and 935 MHz microwave radiation on proliferate ability, colony forming ability and viability of Chinese hamster lung cells, cell line V79. Previously prepared samples of cells in culture were exposed for 1, 2 and 3 hours, both, to 864 MHz microwave field in transversal electromagnetic mode cell (TEM-cell) and to the 935 MHz frequency field in gigahertz transversal electromagnetic mode cell (GTEM-cell). Philips PM 5508 generator connected with signal amplifier generated frequency of 864 MHz, whereas Hellwet Packard HP8657A signal generator was used to generate 935 MHz frequency. Averaged specific absorption rate (SAR) was calculated to be 0.08 for 864 MHz and 0.12 W/kg for 935 MHz. To determine cell growth, V79 samples were plated in concentration of 1x104cells per milliliter nutrient medium. Cells raised in a humified atmosphere at 37 °C in 5% CO2. Cell proliferation was determined by cell counts for each hour of exposure during five post-exposure days. To identify colony-forming ability, cells were cultivated in concentration of 40 cells/ ml medium and incubated as above. Colony forming ability for each hour of exposure was defined by colony counts on the 7th post-exposure day. Trypan blue exclusion test was used to determine cell viability. In comparison to control cell samples, growth curve of 864 MHz irradiated cells showed significant decrease (p<0.05) after 2 and 3 hours of exposure on the 3rd post-exposure day. 935 MHz exposed cell samples showed significant decrease (p<0.05) after 3 hours of exposure on the 3rd post-exposure day. Both, the colony forming ability and viability of 864 MHz and 935 MHz exposed cells did not significantly differ from matched control cell samples. In conclusion, both applied RF/MW fields have shown similar effect on cell culture growth, colony forming ability and cell viability of V79 cell line.
