Vagally mediated pentagastrin cytoprotection
The present study investigated the effect of pentagastrin (PG) on absolute alcohol-induced stomach lesions in rats. MATERIAL and METHODS: The experiment was carried out on male Wistar rats, 200-250 g b.w. Gastric mucosal lesions were induced byoral application of absolute alcohol in dose of 5 ml/kg 30 min before animals were pretreated by PG i.p. (1 mg/kg, 250, 100, 1 ug/kg b.w.). The obtained protective effect has been tested by indomethacin (5 mg/kg s.c.) proglumide (400 mg/kg i.p.), domperidone (25 mg/kg i.p.), atropine (1 mg/kg i.p.) and truncal vagotomy. The influence of the applied substances and animal manipulation on gastric juice secretion and its acidity was also measured using a 4h pylorus ligation. The results were teswted using a Student's t-test. RESULTS: PG in a dose of 1 mg/kg showed a strong protective effect on gastric mucosa (P<0.01). Smaller doses (250 and 100 ug/kg) also have a cytoprotective effect (P<0.05). The smallest dose of 1 ug/kg did not show any protection. Cytoprotective effect was not inhibited by indomethacin, proglumide, domperidone and atropine but the truncal vagotomy inhibited that effect. Vagotomy-, but not atropne- induced inhibition clearly implicate involvement of some other, noncholinergic mechanism. The cytoprotective effect of PG was not related to its effect on gastric juice secretion and to its trophic effect.
