Measurment of autoantibodies against thyroid hormones
The presence of autoantibodies against triiodothyronine (T3) or thyroxine (T4) is known to interfere with RIA of these hormones and it is often in discordance with clinical features. We have measured T3 and T4 binding to serum of 57 patients with thyroid disorders by modification of Allans and Rodriguez-Espinosas methods. The serum was incubated with labelled hormone and counted precipitated radioactivity. The antibody titer was expressed as the percentage of ratio of specific binding to total radioactivity. Normalrange was based on data obtained from 20 healthy subjects and serum free of hormone. T3 binding was 7.9 +/- 0.69% (Mean +/- SD), and for T4 binding 6.3 +/- 0.4%. In group of patients with thyroid disorders % binding T3 was 8.0 +/- 1.73% and for T4 5.8 +/- 1.21%. It was found that T3 binding in 6 patients and T4 binding in 2 patients was higher than 2 SD. We have established them as positive in regard to autoantibodies to T3 and to T4. They should be further detected with immunoelectrophoresis and autoradiorgaphy.
