Sažetak
Non steroidal anti-inflammatory drugs (NSAIDs) have a long clinical use for treatment of various inflammatory diseases like rheumatoid arthritis. However, these drugs are not devoid of serious side effects affecting gastrointestinal and cardiovascular systems. Chemical research of NSAIDs begun in the mid XIX century, with the discovery of salycilic acid derivatives from natural resources, and culminated with the discovery of acethyl salycilic acid over a century ago in Bayer. Since then, many molecules have been designed inspired with the efficacy of "Aspirin" . It took 70 years to discover that NSAIDs inhibit prostaglandin synthesis, and 80 years to isolate the key enzymes that control prostaglandin synthesis, cyclooxygenase I and cyclooxygenase II. Cyclooxygenase I was described as "constitutive" controling normal homeostasis, and cyclooxygenase II as "inflammatory" , involved in ptahological processes in organism. However, new class of cyclooxygenase II specific inhibitors ended, after several years of worlwide use, in withdrawal from market due to increased risk of cardiovascular side effects. The 150 years of research thus was forced to change direction. We generated a new class of compounds, 14 and 15 member ring macrolides, with promising in vitro and in vivo anti-inflammatory activites.
