Modifications of 1-thia-dibenzo[e, h]azulenes at position 8: synthesis and antiinflammatory activity thereof

1-Thia-dibenzo[e, h]azulene derivatives, depicted below by general formula, were found to be new potent molecules with anti-inflammatory activity. One of the main research goals was to investigate the way modifications of different sites at 1-thia-dibenzo[e, h]azulene scaffold affect biological activity. The most obvious and synthetically very interesting modification appeared to be exchange of different heteroatoms (O, S and N) and CH2 group at position 8. Synthetic routes for 8-oxa-, 8-thia-, 8-aza- and 8H-1-thia-dibenzo[e, h]azulene derivatives, respectively, will be presented. In vitro and in vivo data revealed significant differences in anti-inflammatory activity of aforementioned analogues.
