Sažetak
In this work, a comparison of BHK cells growing as natural aggregates or in suspension in batch culture was performed. Cells grow similarly with high viability in all cases. If a small amount of BHK 21 C13 cells (inoculum) are added to the BHK cells forming the aggregates, size of aggregates is becoming bigger. It was confirmed that the aggregate size is opposite with the inoculum concentration. These aggregates can easily retained inside reactors, due to their large size, by sedimentation, while single cels require additional equipment. Scale-up of the BHK cells starts with an initial cultivation at low density in tissue flasks and cell-spin reactors. Bioreactor BR-06 equipped with electronicaly controlled four vertical pole pieces producing rotating magnetic field and automatic oxygen and pH control was used after biomass production in the cell-spin bioreactors. Growth behaviour of BHK 21 cells grown in suspension was mmax.=0.032, viability 92-98 %, population doubling time td/h =12.1 and maximum cell density 2.11x106 cells/ml. When the homogenous mixture of suspended aggregates is reinoculated, no change in growth, viability or any of the measured aggregate characteristics was apparent. Further research are in progress.
