Prepararion and Evaluation of Brush-type Chiral Stationary Phases for Liquid Chromatography Based on 4- or 2-chloro-3, 5-dinitrobenzoic Acid
Chiral stationary phases (CSPs) for liquid chromatography nowadays have a great role in analysis and preparation of enantiomerically pure compounds. Separation of enantiomers on the CSPs is based on formation of short-living diastereomeric complexes between each enantiomer and chiral selector. The enantiomer that forms more stable complex is slowly eluted. Stability of diastereomeric compounds is contributed by non-covalent interactions, such as hydrogen bonding attraction and repulsion, dipole-dipole stacking and p-p interactions. Brush-type or Pirkle-type CSPs are the most widely investigated CSPs and are present in a number of commercial HPLC columns. These CSPs contain a low molecular weight chiral selector covalently bound to the silica gel surface. Recently we have developed a number of original brush-type CSPs and now we presented the novel chiral packing materials for HPLC which are based on 4-chloro-3, 5-dinitrobenzoic acid (CSP I) and 2-chloro-3, 5-dinitrobenzoic acid (CSP II). These new CSPs are proved effective for the resolution of several types of compounds and here we point out the two pharmacologically interesting group: 4-aryl-3, 4-dihydro-2-(1-H)-pyrimidone derivates (DHPM’ s) and b-adrenergic agonists (b-AA) from the class of benzylic b-amino alcohol compounds. DHPM’ s exhibit various important pharmacological properties, e.g. acting as antihypertensive agents, α -1a-adrenergic antagonists, or neuropeptide Y antagonists, and b-AA are known as widely used antiasthmatic drugs.
