Imunohistokemijski aspekti ekspresije proliferacijskih antigena (Ki-67, PCNA), c-erbB-2 i p53 u graničnim tumorima jajnika
During the period from 1991 to 1995 one hundred forty eight maligant ovarian neoplasms were diagnosed, ten of whom were borderline tumors (7 serous and 3 mucinous). Borderline epithelial tumors of the ovary (tumors of low malignat potential) still represent a great problem due to lack of uniform pathohistological criteria and uncertain biological behavior. Therefore, we have examined proliferative kinetics of cells of these tumors. It is well known that these tumors, particularly mucinous, very often contain interspersed areas of well-differentiated as well as atypical cells. Expression of proliferative antigens, Ki-67 and PCNA was studied using Strept AB/AP and Strept AB/HRP immunohistochemical methods after praetretment in microwave owen. The expression of c-erbB-2 and p53 was investigated also by the same methods. Positive nuclear staining reaction for Ki-67 was observed in 4 specimens. The reaction showed mosaic pattern, and was strongly positive in the areas showing epithelial atypia. PCNA nuclear positivity was found in 9 cases, independently of degree of cell differentiation. Positive staining with c-erbB-2 antibody of cell membranous type was observed in 9 tumors, and was mostly expressed in the areas of cytological atypia. Positive nuclear reaction for p53 of mosaic pattern was found in 4 cases, particularly in the areas of atypical epithelium. The expression of examined antibodies in all specimens was strongly positive in areas of cytological atypia and dedifferentiation, except of PCNA which showed positivity also in the areas without proliferative activity. Different expression of proliferative antigens was probably due to their regulation of various phases of the cell cycle. Expression of c-erbB-2 and p53 in investigated tumors indicate their potential agressiveness. Our results of immunohistochemical expression of analyzed antigens may be useful as an additional criteria in morphological evaluation of borderline ovarian neoplasms, and may have a prognostic significance.
