Prevention and suppression of relapsing experimental allergic encephalomyelitis (R-EAE) with a single dose of cyclophosphamide in rats

The effect of a single "high" dose (115 mg/kg) of cyclophosphamide on the development of relapsing form of experimental allergic encephalomyelitis (R-EAE) in susceptible srains of rats was tested. Rats DA and hybrids F1 (AOxDA) were sensitized with bovine brain white matter in complete Freund's adjuvant (CFA). Cyclophosphamide was administered at various intervals of several days before and after encephalitogenic challenge. Our results showed that this schedule of treatment could efficaciously prevent and suppress R-EAE in rats when cyclophosphamide was given before the disease onset, but not earlier than 7 days prior to immunization, and not later than 7 days after sensitization. There were no significant adverse effects among cyclophosphamide treated animals.
