Hematological changes in rats during ochratoxicosis

Ochratoxin A (OA) is a wide spread mycotoxin that could be found in food of plant (max conc. 27.5 mg/kg) and of animal origins (max. conc. 67 micrograms/kg). It could accumulate in kidneys and other tissues taht have higher lipid content. The concentrations of OA in blood of the healthy subjects are between 0.1/14.4 micrograms/L. OA is a potent nephrotoxic, hepatotoxic, gentoxic and carcinogeni agent, and it could interfere with the normal blood coagulation and immune response. We have followed the effects of low doses of OA (120 micrograms/kg b.w.) during 10 and 35 days and of high doses 1200 micrograms/kg b.w. during 10 days (LD 50 is 22-44 mg/kg b.w. for rats). The experiments were performed on the female rats of Wistar stranin (9-10 weeks old ; N = 28). In our experiments we studied following hematological parameters: RBC, WBS, trombocytes, hemoglobin concentration, hematocrite, MCH and differential leukocyte formula. Results revealed statistically significant changes in RBC, after 10 days of treatment with low doses (-12 %), and when higher doses were applied (+ 19 %). Concentration of hemoglobin was in accord with these changes. Following 35 days of treatment with the dose 120 micrograms/kg b.w. we found the significant increase of WBC (+29) but relative decrease in the segmented granulocytes number (-13 %) and increase in the number of lymphocytes (+15 %). The animals that were treated with 1200 micrograms/kg b.w. dose OA showed the statistically significant increase in the number of WBC, relative decrease of segmented granulocytes (-54 %) and increase in lymphocytes count (+19 %). However, the number of trombocytes showed and decreasing trend too.
