Imunohistochemical localisation of ecto-ATPDase in the pig testis

AIM: Localization of ecto ATP diphosphohydrolase (ectoATPDase) in semithin cryosections of pig testis. METHODS: Cryosections of pig testis were analysed for distribution of monoclonal antibody specific for porcine CD 39 lymphoid cell activation antigen, first identified as an ecto ATP diphosphohydrolase, with Daco envision kit. RESULTS: Immunohistochemistry revealed strong staining of the endothelium of the blood vessels, Sertoli cells, spermatogonia, spermatocytes, and round spermatides. Mature forms of spermatogenetic cells i.e. long spermatids and spermatozoa showed no staining CONCLUSIONS: Extracellular ATP has been shown to play important regulatory roles by inducing a wide diversity of biological responses in different tissues. It is rapidly hydrolyzed by various extracellular ATPase and nucleotidase. Sertoli cells under hormonal regulation by FSH and testosterone plays a mayor role in the germ cell differentiation. ATP through activation of P2 purinergic receptors, modulates the biological response of Sertoli cells to FSH. In addition ectoATPDase can generate adenosine which could act on adenosine receptors of Sertoli cells as well as on spermatogenic cells. The precise physiological role of this ectoATPDase in pig testis and its influence on spermatogenesis and sperm cell maturation remains to be elucidated.
