The clinical studies confirmed that moderate alcohol consumption is a beneficial factor in decrease of coronary heart diseases (CHD). Plasma lipoproteins are strongly involved in pathogenesis of atherosclerosis. Low density lipoproteins, LDL, are promoters, while high density lipoproteins, HDL, express the antiatherosclerotic behavior. Therefore, it would be interesting to look at interactions of lipoproteins and alcohol. We have studied the impact of ethanol, in the concentrations which mimicked the beneficial concentrations in the blood stream, on the structure of two human HDL subfractions, HDL2 and HDL3. The fluorescence spectroscopy was applied on unlabeled HDLs and HDLs labeled with membrane probe 6-propionil-2-dimethilaminonaftalen (Prodan). Prodan was used to follow the changes induced by alcohol in the structure of lipid moiety and the modulations in the lipid/protein interface. Its fluorescence behavior is highly sensitive on the micro environment polarity and fluidity. The structure of spectra indicates the partitioning of Prodan in the solvent and in the phospholipids monolayer of HDL. The behavior in the two environments of different polarity and mobility could be followed separately. Changes in spectra observed upon addition of alcohol indicate that the particle structure is pushed to the more fluent loosely ordered phospholipids phase. The fluorescence resonance energy transfer (FRET) from intrinsic protein probe tryptophan, on apoA I to Prodan, in lipid layer, was followed. The spectra indicated changes on lipid/protein interface with increasing alcohol concentration. The proposed model of Prodan behavior in HDL solutions enables interpretation of ethanol impact on the HDL structure.
