Fluorescence microscopy for detecting mycoplasmas in cell cultures

In this study sensibility of florescence microscopy in detection of mycoplasma in cell cultures was established. Twenty cell culture samples were examined by direct method (modified Hayflick, s medium was used for cultivation) and indirect DNA staining method. Both methods revealed the presence of Mycoplasma sp. in 8 samples, and in 12 samples the findings were negative. Mycoplasma hyorhinis ATCC 1798 and Mycoplasma orale ATCC 23714 were positive controls. Only indirect staining method with bis-benzimide-DNA complex revealed both positive controls. Mycoplasma orale ATCC 23714 was detected by direct and indirect methods. Only indirect method showed the growth of positive Mycoplasma hyorhinis ATCC 1798.
