Atypical archaeal seryl-tRNA synthetases recognize tRNASer determinants from all three domains of life
Methane producing archaea Methanococcus jannaschi and Methanococcus maripaludis comprise atypical seryl-tRNA synthetases (SerRSs), which recognise with high efficiency eukaryotic and bacterial tRNASer in additional to their homologous tRNASer and tRNASer substrates. Relative flexibility in tRNA recognition pattern displayed by methanogenic SerRS was shown by aminoacylation and gel mobility shift assays. This indicates the conservation of some serine determinants in all three domains and gives additional support to the existence of a functional connection between archaeal, bacterial and eujaryotic aminoacylation systems. On the other hand, more stringent recognition was observed by yeast SerRS, which does not recognise archaeal tRNA substrates. Homologous M. maripaludis complex of dimeric synthetase with presumably one tRNAser was isolated by gel-filtration chromatography. Complex formation strongly depends on the conformation of tRNA.
